There is no good way to know until you are in Vilanova and speak personally with either the captain or the hospitalero. The schedule varies by the tide and number of passengers.

2 years ago in August the boat left twice the day I was there. Around 7 am and 1 pm.
As stated above it depends on pilgrims and the time of month/year.
There has been several posts from pilgrims saying that when they were there and the numbers of pilgrims were high, an extra boat was arranged by the locals.
Do remember that the locals in general loves pilgrims and will help out as much as possible.

Alternativly call the Oficina de Turismo in pontevedra:
