There are buses all day long from Lisboa, Terminal Rodoviário De Sete Rios.
To get there from the airport you can taxi or take public transport.
Are starting your walk from Fatima or just visiting it?

Lisbon to Santarem along the boardwalks. Then Tagus/Tejo way to Fatima. Then Carmelite route to Coimbra to continue walking or take the train to Porto.
