Where does the Alsa bus leave from in SDC? And do I need a reservation?

Depends on the route but all leave from Estacion de Autobuses . Some might leave from airport but not sure of this.
Reservation : yes better be safe than sorry, especially on the longer distances and when it is a weekend or national holidays/ puentes.
