When can I purchase the online bus tickets from Muxia to Santiago. I need the ticket for May 1st and heard its a holiday. Do the buses still travel that day?

You could probably send a question to Monbus about the bus running on May 1 using their contact form.

You probably won't be able to get a ticket for May 1 before sometime in Mid April or so.
